Seven First Round matches in the J.League Yamazaki Biscuits Levain Cup were played on 9 September.
 
Chiba cruised to a 2-0 victory over Sanuki thanks to goals from Erison and Manato Shinada. Diego Pituca struck the winner in the 77th minute as Nagasaki edged Kochi 1-0.
 
Tokyo V travelled to face Shiga and edged a tight contest 1-0, with Kim Hyun Woo scoring a dramatic winner in the 90+5th minute. Mito, meanwhile, beat Nagano on penalties after the match finished goalless.
 
Yamagata held on to the lead given to them by Kaito Mori's 17th-minute goal to beat Tochigi SC 1-0. Matsumoto drew 3-3 with Shonan before winning the penalty shoot-out 4-3 to knock out their higher-division opponents.
 
Fukushima took the lead at home to Sendai through Tsubasa Nakamura in the 39th minute before an own goal doubled their advantage in the 49th. Sendai pulled one back in the 61st minute, but Kazumasa Shimizu restored Fukushima's two-goal lead six minutes later before Yuki Okada added a fourth in the 70th minute. Sendai struck twice late on, but Fukushima held on for a 4-3 victory to advance to the second round.
